=== Regulation of hCTR1 === hCTR1 is the principal high-affinity Cu(I) importer in human cells. However copper overload results in oxidative damage and cellular toxicity. Consequently, cells maintain strict translational and post-translational control of hCTR1 to prevent copper overload. A key mechanism is copper-stimulated endocytosis of hCTR1. hCTR1 is rapidly internalized from the plasma membrane after exposure to elevated copper Copper-triggered endocytosis of hCTR1 is clathrin dependent and reversible. When extracellular Cu is depleted or reduced, internalized hCTR1 is re-routed to restore copper uptake capacity. But prolonged high copper concentration can also target internalized hCTR1 for lysosomal degradation, leading to a net decrease in hCTR1 protein levels. == Physiology == Free fatty acids (FFAs) are liberated from lipoproteins by lipoprotein lipase (LPL) and enter the adipocyte, where they are reassembled into triglycerides by esterifying them onto glycerol. Human fat tissue contains from 61% to 94% lipids. Lean and obese individuals tend towards the low and high ends of this range, respectively. There is a constant flux of FFAs entering and leaving adipose tissue. The net direction of this flux is controlled by insulin and leptin—if insulin is elevated, then there is a net inward flux of FFA, and only when insulin is low can FFA leave adipose tissue. Insulin secretion is stimulated by high blood sugar, which results from consuming carbohydrates. In humans, lipolysis (hydrolysis of triglycerides into free fatty acids) is controlled through the balanced control of lipolytic B-adrenergic receptors and a2A-adrenergic receptor-mediated antilipolysis. Fat cells have an important physiological role in maintaining triglyceride and free fatty acid levels, as well as determining insulin resistance. Abdominal fat has a different metabolic profile—being more prone to induce insulin resistance. === Other forms === The anamorphic form of P. semilanceata is an asexual stage in the fungus's life cycle involved in the development of mitotic diaspores (conidia). In culture, grown in a petri dish, the fungus forms a white to pale orange cottony or felt-like mat of mycelia. The conidia formed are straight to curved, measuring 2.0–8.0 by 1.1–2.0 μm, and may contain one to several small intracellular droplets. Although little is known of the anamorphic stage of P. semilanceata beyond the confines of laboratory culture, in general, the morphology of the asexual structures may be used as classical characters in phylogenetic analyses to help understand the evolutionary relationships between related groups of fungi. Scottish mycologist Roy Watling described sequestrate (truffle-like) or secotioid versions of P. semilanceata he found growing in association with regular fruit bodies. These versions had elongated caps, 20–22 cm (7.9–8.7 in) long and 0.8–1 cm (0.3–0.4 in) wide at the base, with the inward curved margins closely hugging the stipe from the development of membranous flanges. Their gills were narrow, closely crowded together, and anastomosed (fused together in a vein-like network). The color of the gills was sepia with a brownish vinaceous (red wine-colored) cast, and a white margin. The stipes of the fruit bodies were 5–6 cm (2.0–2.4 in) long by 0.1–0.3 cm (0.04–0.12 in) thick, with about 2 cm (0.8 in) of stipe length covered by the extended cap. It is also a partial agonist of the serotonin 5-HT1A receptor (EC50 = 2,300 nM; Emax = 74.7%) and of the serotonin 5-HT1D receptor (EC50 = 262 nM; Emax = 57.1%). Conversely, its activities at various other targets, such as various other serotonin receptors as well as adrenergic and dopamine receptors, are much less potent. TAAR1 agonism is known to reduce the firing rate of dopaminergic neurons. The inhibitory effects of TAAR1 agonists on dopaminergic neurotransmission are most pronounced in hyperdopaminergic states. Ulotaront decreases basal locomotor activity in rodents and this effect was absent in TAAR1 knockout mice. It prevented the hyperlocomotion induced by the NMDA receptor antagonist phencyclidine (PCP). Conversely, ulotaront did not affect dextroamphetamine-induced hyperlocomotion.
